Anti - Inflammatory Activity of Quercetin and Kaempferol Is Limited to Tlr4 Stimulation in Monocytes

Flavonoids are known to exert anti-inflammatory activity but their mechanism at receptor level and comparative potencies less studied. Objective of the study is evaluate, rank order few flavonoids for cytokine inhibitory in monocytes (THP-1) explore signature among toll-like receptors (TLRs). We tested potential inhibit LPS induced cytokines IL-6, TNF alpha human monocyte cell line THP-1 mouse macrophage using Enzyme linked immunosorbent assay (ELISA) assays. Quercetin Kaempferol reduced secretion lines. demonstrated dose dependent inhibition IL-6 alpha. IC50 6.9µM whereas 14.3µM reduction. In THP-1, NF-kB measured secreted embryonic alkaline phosphatase (SEAP) reporter with different activation. TLR4 stimulated was inhibited by line, while these did not impact signaling other TLR ligands Pam3CSK4 (TLR1/2) FLS (TLR2/6), Flagellin (TLR5) R848 (TLR7/8). I. modulate when TLR4, TLR1/2, TLR2/6, TLR5 TLR7/8 Monocytes. Understanding profile against Toll will help design better inhibitor which can alleviate inflammatory diseases
